Provisional schedule
1.      FRET - Zeiss Cell Observer SD spinning disk confocal & MBF Spinning Disk Confocal*

Fixed Mammalian cells expressing a C5V will be used as a model for quantifying FRET. Images will be acquired to quantify sensitised emission FRET as well as acceptor photo bleaching FRET.

2.      FRAP -  Olympus FV1000

Cells expressing cytosolic or membrane targeted fluorescent protein will be used to quantify mobility in the cytosol and in membranes.

3.      Spectral analysis - Leica TCS-SP5 X white-light confocal & Leica MP TCS-SP5*

Spectral separation of similar fluorescent proteins. Fixed cells expressing YFP, GFP or YFP and GFP.

4.      Ratio-imaging of Biosensors - Ratio-Imaging Workstation* & PTI EasyRatioPro

Dual-wavelength dyes and biosensors are increasing in number as reporters for dynamic cellular events such as changes in ion concentration, protein phosphorylation and protease activity. 

5.   Widefield Deconvolution - Deconvolution Workstation* & Nikon Ti-E

A comparison will be made using widefield deconvolution and structured illumination of live yeast expressing GFP and RFP.

6.     Image Processing and Analysis with ImageJ - located in the CSU training suite HSC 2D8

A hands-on introduction to ImageJ covering installation, upgrading, plugins and macros, routine processing and analysis and an introduction to writing macros to automate and write custom functions.

7. Live cell confocal. Perkin Elmer Ultraview
