The Microscope Part 2: Different 'scopes for different folks. - T. J. Collins Fluorescence Microscopy has undergone a revolution in recent years with the development of cheaper, more powerful light sources; the increased processing power of personal computers and the development of novel fluorescent reagents. Along with these developments, new microscope modalities have emerged. The traditional wide-field microscope has been joined by single-point scanning confocal and multi-point scanning confocal microscopes. The 100-year-old diffraction limit of resolution is also being challenged with hyper-resolution microscopes. This seminar will compare these different microscopy modalities and how they work; in addition I will discuss the advantages and disadvantages associated with each approach. |
